PTSD Rarely Stands Alone; Why Understanding Comorbidity Matters

In the clinical and research worlds, post-traumatic stress disorder (PTSD) is often treated as a singular diagnosis, but the reality is that PTSD rarely occurs in isolation. Comorbid conditions such as alcohol use disorder (AUD), depression, anxiety, and traumatic brain injury frequently accompany PTSD, complicating diagnosis, clouding treatment paths, and worsening patient outcomes.

Despite the high prevalence of these overlapping conditions, biological research has traditionally treated them in silos. That approach leaves critical questions unanswered: Why are some individuals more vulnerable to both PTSD and substance use? Are there shared molecular mechanisms driving comorbidity? And what does this mean for how we diagnose and treat people more effectively?

This gap in understanding represents both a challenge and an opportunity. Polaris Genomics, alongside academic research partners, is working to close it.

By integrating multi-omics analysis – spanning genetics, transcriptomics, and proteomics –  Polaris and its research partners are pursuing the biomarkers that underlie PTSD and its comorbidities. Their approach explores not just the presence of symptoms, but the molecular interplay that may make one condition exacerbate another.

At the center of this effort is Polaris’ platform, which enables researchers to conduct more precise, blood-based analyses of comorbid conditions. This offers a window into brain-related pathology that is both scalable and clinically actionable. In the future, clinicians will no longer be limited to retrospective analysis or self-reported symptoms; they can begin to track PTSD as a biological condition with defined and measurable molecular characteristics.

The potential of this work is significant. Not only could it lead to earlier, more objective diagnostic tools, but it also lays the groundwork for treatment tailored to patients living with multiple, interacting mental health challenges. That includes the many veterans and trauma survivors for whom traditional treatment protocols have failed.

As we observe National PTSD Awareness Month, it’s important to widen the lens. PTSD is often not an isolated condition, and our research frameworks shouldn’t treat it that way. Polaris Genomics is advancing biologically grounded research that can turn complex mental health profiles into clearer, more personalized paths forward.
